เรื่องราวว่าด้วยลูกขุนทั้ง 12 คนที่ได้รับมอบหมายให้มาตัดสินคดีของผู้ต้องหาที่เป็นเด็กชายคนหนึ่งที่ถูกกล่าวหาว่ากระทำปิตุฆาต(ฆ่าพ่อตัวเอง)ว่าผิดจริงหรือไม่? โดยมีกฎว่าคำตัดสินขอลูกขุนทุกคนต้องออกมาเป็นเอกฉันท์เท่านั้นถึงจะตัดสินคดีและลูกขุนทุกคนสามารถออกจากห้องพิจารณาคดีได้ แต่ถ้าเสียงไม่เป็นเอกฉันท์ก็ต้องถกกันไปเรื่อยๆจนกว่าลูกขุนทุกคนจะเห็นพ้องกัน ซึ่งตอนแรกลูกขุนส่วนใหญ่ก็ฟังธงกันไปเรียบร้อยแล้วว่าเด็กชายผิดจริงแน่นอนเมื่อดูจากหลักฐานมากมายในชั้นศาล…แต่ผลโหวตกลับออกมาเป็น ผิดจริง 11 เสียง ไม่ผิดจริง 1 เสียง เพราะหนึ่งในลูกขุนทั้ง 12 คน ลูกขุนหมายเลข 8 (Henry Fonda) เห็นว่ามันยังเร็วเกินไปที่จะตัดสินประหารชีวิตคนโดยไม่ไตร่ตรองเสียก่อน ทำให้ลูกขุนทั้ง 11 คนจำต้องหาวิธีพูดโน้มน้าวลูกขุนหมายเลข 8 คนนี้ให้ได้ว่าผู้ต้องหา (เด็กชาย) ผิดจริงตามที่พวกเขาคิด……ก่อนที่พวกเขาจะพบว่า ยิ่งถกกันมากเท่าไหร่ พยานหลักฐานต่างๆก็ยิ่งมีความขัดแย้งกันเองมากขึ้นเท่านั้น สรุปแล้วเด็กชายผิดจริงรึไม่
